This paper presents a method to estimate the biomass of Spirulina cultivated on solid medium with sugarcane bagasse as a support, in view of the difficulty in determining biomass concentrations in bioprocesses, particularly those conducted in semi-solid or solid media. The genus Spirulina of the family Oscillatoriaceae comprises the group of multicellular filamentous cyanobacteria (blue-green microalgae). Spirulina is used as fish feed in aquaculture, as a food supplement, a source of vitamins, pigments, antioxidants and fatty acids. Bioconversion of biomass: a case study of ligno-cellulosics bioconversions in solid state fermentation BABT

Lignocellulosic residues obtained from crops cultivation form useful sources to be used as substrate for bioconversion processes. Sugarcane bagasse, which is a complex substrate obtained from the processing of sugar cane, is an important biomass among such sources. Due to its abundant availability, it can serve as an ideal substrate for microbial processes for the production of value added products. This paper reviews recent developments on biological processes developed on production of various products in solid state fermentation using sugarcane bagasse as the substrate and describes production of protein enriched feed, enzymes, amino acid, organic acids and compounds of pharmaceutical importance, etc. through microbial means.

Cellulase production by thermophilic Bacillus sp: SMIA-2 and its detergent compatibility Electron. J. Biotechnol.

Background This paper reports the production of cellulase by thermophilic Bacillus sp. SMIA-2 using sugarcane bagasse and corn steep liquor as substrates. Some biochemical properties of the enzyme were also assessed for the purposes of exploiting its potential in the detergent industry, as well as other suitable applications. Results Bacillus sp. produced cellulases when cultivated at 50°C in liquid cultures containing sugarcane bagasse and corn steep liquor. Maximum avicelase (0.83 U mL-1) and CMCase (0.29 U mL-1) activities were reached in 120 h and 168 h of culturing time, respectively. Biocatalyst consisting of immobilized cellulases for hydrolysis of pretreated sugarcane bagasse by organosolv process. The combination with easily hydrolyzable cellulose allows obtaining highly pure glucose and the reuse of enzymes. The immobilized enzyme represents most of the solid recovered at the end of the hydrolysis because almost all the solid substrate added to the reactor is solubilized.

Extracellular production of avicelase by the thermophilic soil bacterium Bacillus sp. SMIA-2 Biological Sciences

Nowadays, the isolation of new bacterial strains that produce enzymes with novel properties is a subject of great relevance to the scientific community. This study, in order to search for producers of new cellulase strains, investigated the avicelase production by thermophilic Bacillus sp. strain SMIA-2. The best avicelase activity was observed in a culture medium containing 0.5% (w v-1) avicel and 0.5% (w v-1) corn steep liquor with initial pH 7.5-8.0 incubated at 50oC. When avicel was replaced in the medium by the treated sugarcane bagasse (0.5%, w v-1) the avicelase activity levels were not affected. Filamentous fungi and media for cellulase production in solid state cultures BJM

Cellulase production was evaluated in two reference strains (T. reesei Rut-C30 and T. reesei QM9414), two strains isolated from a sugarcane cultivation area (Trichoderma sp. IPT778 and T. harzianum rifai IPT821) and one strain isolated in a program for biodiversity preservation in São Paulo state (Myceliophthora thermophila M77). Solid state cultures were performed using sugarcane bagasse (C), wheat bran (W) and/or soybean bran (S). The highest FPA was 10.6 U/gdm for M77 in SC (10:90) at 80% moisture, which was 4.4 times higher than production in pure W. Production of cellulases by Penicillium oxalicum through solid state fermentation using agroindustrial substrates Biological Sciences

 The purpose of this study was to define the factors that influence the production of cellulases by Penicillium oxalicum, a strain obtained from a leaf-cutting ant colony and identified based on the ITS gene. The experimental design included solid state fermentation using sugarcane bagasse and lignocellulosic sorghum as the lignocellulosic substrate. The variables were analyzed using a 25-1 fractional factorial design, with three replicates on the central point. All the variables analyzed influenced the production of at least one of the three cellulose types analyzed. Shiitake (lentinula edodes) production on a sterilized bagasse substrate enriched with rice bran and sugarcane molasses BJM

This investigation was performed to evaluate the biological efficiency (BE), mean mushroom weight (MMW), mean number of mushroom (MNM) and mushroom quality of Shiitake [ Lentinula edodes (Berk.) Pegler] when grown on a sterilized substrate composed by sugarcane bagasse enriched with rice bran and sugarcane molasses. The proportions of rice bran were 0, 15, 20, 25 and 30% (dry weight/dry weight of bagasse); and the concentrations of sugarcane molasses were 0, 30 and 60 g/kg (dry weight/dry weight of bagasse plus rice bran). Supplementation of sugarcane bagasse with rice bran and sugarcane molasses for shiitake (Lentinula edodes) spawn production BJM

The purpose of this study was to assess the myceliation rate, mycelial vigor and "estimated biomass" of Lentinula edodes (Berk.) Pegler, grown on a sugarcane bagasse substrate enriched with rice bran and sugarcane molasses for spawn production. The proportions of rice bran used were 0, 10, 15, 20, 25, 30 and 40% (dry weight/dry weight of bagasse) and the sugarcane molasses concentrations tested were 0, 10, 20, 30, 40, 50 and 60 g/kg (dry weight/dry weight of bagasse plus rice bran). The myceliation rate was decreased by the addition of the higher quantities of rice bran. The 25 and 30% rice bran proportions induced the highest stimulation of mycelial vigor. Thermotolerant and mesophylic fungi from sugarcane bagasse and their prospection for biomass-degrading enzyme production BJM

Nineteen fungi and seven yeast strains were isolated from sugarcane bagasse piles from an alcohol plant located at Brazilian Cerrado and identified up to species level on the basis of the gene sequencing of 5.8S-ITS and 26S ribosomal DNA regions. Four species were identified: Kluyveromyces marxianus, Aspergillus niger, Aspergillus sydowii and Aspergillus fumigatus, and the isolates were screened for the production of key enzymes in the saccharification of lignocellulosic material. Among them, three strains were selected as good producers of hemicellulolitic enzymes: A. niger (SBCM3), A. sydowii (SBCM7) and A. fumigatus (SBC4). Xylanase production by Trichoderma harzianum rifai by solid state fermentation on sugarcane bagasse BJM

Sugarcane bagasse was used as substrate for xylanase production by means of a strain of Trichoderma harzianum Rifai isolated from decaying Aspidosperma sp. (peroba) wood. The bagasse was washed, dried, milled and wetted with minimal salts medium and the cultures grown at 28 ± 2ºC for 7 days. Two extraction methods were tested for enzyme recovery: (A) Tween 80, 0.1% (v/v), in physiological saline, and (B) 50mM sodium acetate buffer, pH 5.0, under agitation (180rpm) for 15, 30 and 60min. After a single extraction, both extraction methods recovered an average of 15U/ml of xylanase activity, independent on the time of shaking.
